Quantitative RT–PCR

Quantitative PCR is used at PsychoGenics to quickly examine gene expression changes on mRNA levels. Expression level of a gene of interest is normalized to the mean values of expression level of housekeeping genes to provide accurate quantification.

Figure 1: Between groups comparison of effects of genotype on nitric oxide synthase (nNOS) transcripts in striatum and cortex of WT, R6/2, and z_Q175 KI mice models of Huntington’s disease. nNOS mRNA expression is significantly down-regulated in the (A) striatum (54%) and (B) cortex (40%) of R6/2 mice at 3 months of age [unpaired t-test, ****p<0.0001]. Data are derived from 12 wildtype (WT) mice (n=6 male, n=6 female) and 12 R6/2 mice (n=6 male, n=6 female). A more modest reduction (19%) of nNOS transcript levels was observed in the (C) striatum of 11.5 month old Q175 mice (unpaired t-test, **p<0.01), whereas the (D)cortex was not affected (p>0.05; ns, not significant). Data are derived from 12 WT mice (n=6 male, n=6 female) and 11 Q175 mice (n=6 male, n=5 female).
